Nine and producers Easy Tiger have confirmed the news.
The show, which transformed Mudgee into the fictional town of Whyhope, followed Hugh Knight, played by Rodger Corser, a heart surgeon, who is punished and is put on probation and forced to work in the country for a year as a G.P.
It debuted in 2016 with production back and forth from Mudgee ever since, putting the national spotlight on the town.

Channel 9's Head of Drama, Andy Ryan and Easy Tiger's Founder and Producer, Ian Collie provided a statement regarding the show's cancellation.
"For five incredible seasons, Australia has shared in the journey of everybody's favourite bad boy doctor, Hugh Knight, his fabulously flawed family and the beautiful town of Whyhope," they said.
"But all journeys have a destination, and after five seasons it's finally crunch time in Hugh's roller-coaster search for redemption and love. We are thrilled to bring the story to a fitting climax in a way that honours the characters and will surprise and delight the audience.
"Doctor Doctor has been a domestic and international success, as well as a showcase of outstanding Australian talent. Channel 9 and Easy Tiger are immensely proud of what Doctor Doctor has achieved. It has been a privilege to entertain our loyal viewers, as well as collaborate with our magnificent cast and crew for five seasons".
Speaking in 2018, lead actor Rodger Corser explained why he thought the show was so popular.
"It has that mix of comedy and drama, we've got heart and warmth," he said.
"We don't discount how great the show looks either, it's a real credit to all of the crew. But also, Mudgee is a big star of the show, there's a reason we chose this location. We can go and shoot at a winery or a mine or in Market Street with the famous clock in view. Not to mention all the beautiful buildings around."
