A Nick Karydis five-wicket haul saw Souths rolled for just 60, and first innings points were secured with minimum fuss.
A declaration saw Souths sent in again and they finished the day at 4/54, just the 26 runs ahead at stumps.
Rugby opening bowler Lawson Shepherd took the day's first wicket in just the fourth over and it was a procession from there.
Karydis did the bulk of the damage and finished with the outstanding figures of 5/8 from 8.3 overs while Shepherd finished with 2/13.
Matt Naden banged a quick 47 to have first innings points wrapped up with a minimum of fuss and his declared at 4/88.
Zac Murphy then bagged two late wickets to have Souths in all sorts of trouble heading into day two.
There was also wickets galore at Lady Cutler 1 where Colts proved far too strong for Macquarie.
Colts are also on track for an outright win after rolling the Blues for just 61, with Bailey Collins taking 3/18.
Colts went hard with the bat to build a strong lead and while there was no dominant effort with the bat a number of contributions helped the men in red post 150.
Luke Patis bagged four wickets but his side will need something special when it resumes at 1/0 next Saturday.
The other match is a little more in the balance, but CYMS will be hopeful two late wickets will give them the advantage in the race for third.
CYMS and Newtown are both level on 44 points and the Cougars posted 193 on Saturday, with Connor Watts' 65 leading the way.
Newtown had 17 overs to survive with the bat and didn't get through unscathed, finishing at 2/38.
In the RSL-Kelly Cup third grade competition, Saturday was a momentous day for RSL-Colts United's Geoff Wheeler.
Wheeler took 5/16 from eight overs against Newtown Gold and that haul sent him to the top of Colts' all-time leading wicket-takers chart with 525 scalps.
Gold was rolled for 67 in reply to Colts total of 98.
Top two Newtown Demons and CYMS White both won again.
After David Artery's 4/17 restricted CYMS Green to 9/163, Mick Daly (75) and Bevan Charlton-White (51 not out) fired the Demons to a strong eight-wicket win.
CYMS White downed South Lemons despite a five-wicket haul from Jeff Shanks.
Shanks took 5/41 but Zac Williams (45) and Beau McIntosh helped CYMS White post 203.
McIntosh then took three wickets as Lemons were knocked over for 159.
Steve Merritt's six wickets fired Rugby to victory over Macquarie.
Glenn Healey's 59 not out led Rugby to a total of 5/164 and then Merritt took 6/20 as Macquarie was knocked over for 141.
RSL-Colts Coolbaggie scored a tight win over Souths Limes in the final match of the round.
Coolbaggie posted 155 and then a disciplined and consistent bowling effort saw Limes restricted to 8/136 in reply.
