Dubbo MP Dugald Saunders announced the start of the project at the intersection of Fitzroy Street and the Mitchell Highway, which is being undertaken to improve safety and reduce travel times.
He said it would reduce congestion, especially during peak times.
"Congestion is a growing problem for Dubbo as more people come to live and work here," Mr Saunders said.
"That's why we're getting on with delivering key intersection upgrades at the major pinch points."
"This section of the Mitchell Highway, in the centre of Dubbo, is a key route for residents, commuters and visitors and a vital route between Dubbo and Orange."
The upgrade will include widening the highway, building a new pedestrian crossing, and safety features including upgraded street lighting and accessible ramps, Mr Saunders said.
Early work will continue until October 25 on weekdays between 7pm and 12am, weather permitting.
Shoulder closures and lane reductions will be in place during day work including 40km/h speed limits.
Early work includes the removal of trees and establishing a site compound to enable the future widening work.
A resident of the neighbourhood had earlier spoken of his disappointment at the loss of an avenue of trees along Fitzroy Street, part of the early work.
Although offset trees were planted in Elston Park, Mark Gardner called for trees to be replanted along Fitzroy Street to provide shade and habitat.
