The bridge on the Newell Highway will be closed to traffic between 7pm on May 4, and 6am on May 5, to allow for spray sealing.
During the overnight closure, traffic including heavy vehicles will detour through Thompson Street, Bunglegumbie Road and Troy Bridge Road.
Motorists are also being advised to use the LH Ford Bridge to get across the Macquarie River.
The majority of the works program will be conducted between 7pm and 6am on May 2, May 3, May 5 and May 6, weather permitting.
Two lanes of the bridge will be closed on the four nights, leaving one northbound lane and one southbound lane open.
This week state Member for Dubbo and NSW Deputy Premier Troy Grant announced the pending start to the project and changed traffic arrangements.
The state government is paying for the construction of a "safer wet weather road surface" on the bridge with the work to be carried out by the council on behalf of Roads and Maritime Services.
The Emile Serisier Bridge was built in 1986 and opened to traffic in 1987.
"The planned work includes resurfacing of the bridge, removing some of the existing road surface and applying new asphalt," Mr Grant said.
"The bridge is a 90-metre-long, low-level concrete bridge built in 1986 and the surface is in need of an upgrade."
