Souths dominated day one of the Whitney Cup clash last week, rolling the Cougars on a rain-affected pitch for 99 before posting 5-200 at stumps.
With the lead already over 100 and 75 overs still to be played out the Hornets are well placed to score an outright win and move into top spot on the ladder but Mitch Bower and his men will be taking nothing for granted.
More batsman-friendly conditions are also expected on Saturday, making Souths task that bit more difficult.
Bower hinted last week his side would declare at the start of play, giving his bowlers the maximum amount of time possible to dismiss CYMS a second time.
However, the option is also there for the Hornets to spend longer at the crease and go for 300 and the maximum batting points.
Mat Finlay remains at the crease, unbeaten on 49, while Tim Berry's return cancels out the unavailability of Greg Rummans.
Next door at No.2 Oval, the RSL-Colts bowlers will look to follow on from the good work done by their batsmen last week.
The defending premiers posted 9-286 against Rugby last week.
Rugby have only scored that many runs twice this season and on both those occasions captain Jordan Moran posted big runs.
Moran scored 111 when his side made 9-332 against CYMS and he followed that up the next match with 82 of his team's 303.
Apart from those two innings, Rugby have only passed 200 one other occasion this season meaning the pressure is well and truly on the men in sky blue.
As well as Moran, Rugby possess a lineup featuring Nathan Munro and James O'Brien while tailender Jacob Bruce has proved multiple times this season he is more than capable of scoring big runs.
Colts will be without Marty Jeffrey but are boosted by the return of Tim Cox, to boost and attack already featuring Ed Morrish, Ben Semmler, youngster Tom Atlee and spinners Greg Buckley and Wes Giddings.
Both matches resume at 1pm on Saturday.
