Rugby shot to the top of the standings after the lopsided win over CYMS while Souths sits second having downed RSL-Colts in round one.
A win this time out would likely put Rugby even further in front and put the men in sky blue in right in the finals frame early in the summer.
However, Rugby will be without Adrian Carey, who took eight wickets last weekend against CYMS, while Souths will enjoy a much more strong lineup this time out.
Opening weekend centurion Josh Williams retains his place in the side while captain and former NSW Country player Mitch Bower returns after missing round one.
Bower’s battle with Rugby quicks Ben Patterson, James O’Brien and Dan Horrocks, all who impressed against the Cougars, could go a long way to deciding the contest.
O’Brien will also be looking to make an impact with the bat after starting his summer in fine form when blasting 123 not out against CYMS.
The way the pitch plays after rain late in the week could also be a key component in the fixture.
Jacob Hill returns to the Rugby lineup in place of Carey after missing round one with that being the only change.
Day one play begins at 1pm at No. 2 Oval.
