The head of the investigation, homicide Detective Inspector Gary Jubelin, arrived at the Bonny Hills semi-rural property a few minutes after Mr Spedding was seen leaving in his work van about 8.30am.
Inspector Jubelin walked around the property with a colleague for several minutes before leaving.
Police have repeatedly stressed the 63-year-old is not a suspect but a "person of interest" in relation to the suspected abduction of the toddler who vanished from his grandmother's Kendall home almost six months ago.
Mr Speeding is a former businessman and resident of Wellington.
The local tradesman has come under intense media scrutiny after police spent 48 hours searching his Bonny Hills property and Laurieton office in January.
A number of items were seized including cars, computer equipment and a single mattress - all of which were sent for forensic testing.
Police refuse to talk about the results.
No charges have been laid and two of Mr Spedding's children maintain their father has done nothing wrong and is distraught by the continued media attention.
Up to 30 officers will continue to scour a dense and muddy bush track about six kilometres from Mr Spedding's home and 20 kilometres from where the toddler disappeared in Kendall.
Police said the search came after fresh information was received in relation to William's disappearance and was expected to continue until Wednesday afternoon.
William was wearing a Spider-Man suit and playing with his little sister when he vanished from his grandmother's garden on September 12, 2014.
