Dubbo City Council manager of environmental control Debbie Archer said rangers were called to the Newell Highway near Dubbo Golf Course about 10.30pm following reports of the crash.
"One goat died as a result of the accident and about 70 of them got out," she said.
"Our rangers managed to round some up in the early hours of the morning but there are still about 50 missing.
"It was very dark and obviously very difficult to find many of them."
Ms Archer has urged members of the public to report any goat spottings.
"Our primary concern is that they pose a traffic hazard," she said.
"They may be in groups or there may be the odd one running around by itself.
"Please let council know if you see them.
"The sooner we know where they are the sooner we can get them."
A spokesperson for Orana Local Area Command said the truck overturned after it was hit by a gust of wind as it turned from Obley Road onto the Newell Highway just after 10pm on Monday.
"The vehicle did one full rotation and came to rest on its wheels, and the goats escaped," he said.
Paramedics treated the 65-year-old male driver and 19-year-old passenger at the scene before they were taken to Dubbo Hospital in a stable condition, a NSW Ambulance spokesperson said.
