Police officers alerted firefighters after finding a Bathurst Street building on fire about 8pm.
Occupants in the area were evacuated and no one was injured.
The fire destroyed the original building and an adjoining building.
Crews from Brewarrina and Bourke extinguished the blaze.
A spokesperson for the Rural Fire Service (RFS) Far West Zone said the buildings destroyed were the historic main street cafe and the old pharmacy.
Bathurst Street between Belmore and Sandon Street remains closed with a diversion via Bourke Street and Darling Street.
