The world No.116, and last Australian in the draw, was beaten 6-7 (4-7) 7-6 (11-9) 3-6 in just under three hours nine minutes by Serbia's Laslo Djere in round-of-16 action on Wednesday.
Richard Gasquet will face Djere in the last eight after the Frenchman defeated American Steve Jonson 6-7 (3-7) 6-3 6-3 while Austrian wildcard Dominic Thiem fell in just 80 minutes to Britain's Jack Draper, 6-1 6-4.
Thiem, playing in his first tournament on American soil since winning the 2020 US Open, had reached the third round after top-seeded Grigor Dimitrov of Bulgaria retired in round two because of dizziness.
"I was really good behind serve, really solid," 20-year-old Draper, who reached his third quarter-final of the season after Eastbourne and Montreal, said.
"In the second set it was tough because he was coming up with some amazing shots. It was a real honour to be on court with Dominic. He's an amazing player."
Draper's reward is a clash with defending champion Ilya Ivashka of Belarus or Switzerland's Marc-Andrea Huesler.
Botic Van de Zandschulp cruised into the quarters by beating Spain's Jaume Munar 6-3 6-3.
The second-seeded Dutchman advanced to play 10th-seeded Benjamin Bonzi of France, a 7-5 6-2 winner over Brazil's Thiago Monteiro.
Fourth-seeded American Maxime Cressy saw off No.14 seed Lorenzo Sonego of Italy 6-7 (4-7) 6-2 7-6 (7-3) and moved on to face France's Adrian Mannarino, who cruised past eighth-seeded Spaniard Albert Ramos-Vinolas 6-3 6-3.
