A teenage boy and a man died, and three people were seriously injured, after the car they were travelling in hit a tree near Mendooran on Saturday afternoon, police said.
Hours earlier, a driver died in Dubbo Hospital after his vehicle and a truck collided at Ballimore on Friday evening.
And shortly before 4pm on Friday a 72-year-old man died in a single-vehicle crash on Henry Parkes Way east of Manildra near Orange.
Their deaths add to the state's road toll, which was at 165 fatalities by Thursday's end.
On Monday police will continue investigating the crash near Mendooran, about 80 kilometres north-east of Dubbo.
Emergency services were called to the Castlereagh Highway at Merrygoen, about 10km east of Mendooran, at 3.20pm on Saturday, after a 2005-model Mitsubishi 380 sedan travelling west left the road and hit a tree.
Three rescue helicopters with critical care medical teams on board were part of the multi-agency response.
The driver and four passengers were trapped in the vehicle and had to be released by emergency services, police said.
The front-seat passenger, a 14-year-old boy, and a 25-year-old man, who was in the back seat, died at the scene.
Police confirmed the 25-year-old man was from Gulgong, while the others were all from Menah.
The driver, a 37-year-old woman, was airlifted to Westmead Hospital with arm fractures, police said.
A 13-year-old boy was airlifted to The Children's Hospital at Westmead with multiple leg injuries, and a 17-year-old girl was airlifted to John Hunter Hospital also suffering leg fractures.
The Westpac Rescue Helicopter Service reported on Saturday night occupants had been airlifted by two helicopters to Westmead and Westmead Children's Hospital, and the Westpac Rescue Helicopter was airlifting a 16-year-old female to John Hunter Hospital with multiple injuries in a critical condition arriving at 8pm.
The crash closed the Castlereagh Highway between Gilgandra and Dunedoo in both directions for some time.
On Friday about 6pm emergency services were called to the intersection of the Golden Highway and Ballimore Road at Ballimore, east of Dubbo, following reports a Toyota LandCruiser and a truck had collided.
The Toyota driver, a 35-year-old man, was found critically injured after being ejected from the vehicle while the passenger, a six-year-old girl was also injured, police said.
Both were treated by paramedics and taken to Dubbo Hospital.
The man was placed on life support, however, died shortly after 1am on Saturday, police said. The girl was airlifted to John Hunter Hospital, where she remained in a serious condition.
The truck driver, a 46-year-old man, underwent mandatory testing.
An investigation has started and a report will be prepared for the coroner.

Officers from the Central West Police District are investigating the single-vehicle crash along Henry Parkes Way.
Police have been told a Honda Civic was travelling west when it veered off the road and crashed into a tree.
The driver, a 72-year-old man, was treated by NSW Ambulance paramedics but died at the scene.
