The crash occurred at 7.08am on Monday with a b-double rolling and spilling tens of thousands of litres of fuel.
The highway is closed in both directions at Elong Elong, east of Dubbo between Cobbora and Muronbung roads.
Diversions are in place with westbound motorists advised to take Cobbora Road and Muronbung Road to re-join the Golden Highway. Eastbound motorists should do the same but in reverse.
This diversion is suitable for all vehicles.
"The highway is closed until further notice due to fuel contamination of the soil," Live Traffic said.
Australian Community Media understands that this section of the highway may remain closed until the end of January.
