The ladder leaders overcame a disappointing start on day one of the clash, dismissing Rugby for 63 after posting just 154 themselves.
But day two will see the Tigers going for the jugular, knowing an outright victory would put them in pole position for the minor premiership and through to the 2018-19 season final.
"It'd be huge - it'd put us up there with a nice gap at the top of the ladder," said Steve Skinner who, with 4-6 from 8.2 overs, was a key component in last weekend's dominance.

The Tigers will be boosted by the return of captain and right-arm quick Mat Skinner.
Top order batsman Charlie Kempston admitted a win in Friday night's McDonalds Megahit grand final would be a welcome confidence boost as well.

The Tigers will look to extend their total - Steve declined to predict their target - before sending Rugby in against quicks Ben Patterson, Trent Smith and Mat Skinner.
Young "work horses" Lee Price and Jesse Spang will go to work in the middle overs, the latter trusted to fill the shoes of Tom Barber.
Steve was the pick of Newtown's bowlers last week, and said that while the "ball's been coming out pretty well all year", he never expected figures like 4-6.
He doubts he'll have the same sort of luck on Saturday, saying Rugby's defensive approach had helped the Tigers quicks get "a roll on".

Campbell Rose (11) and Nick Karydis (22) gave Rugby's chase the best possible start, moving to 40 before a wicket fell.
But the openers' falls triggered a sensational collapse, with Baye Wallace (0), Koda Sissian (0), Peter Karydis (0) all falling in quick succession.
Rugby captain Nathan Munro (16) was the only other batsman to make double digits.
"They let us be a bit aggressive with the fields we were setting and where I was able to bowl," Steve said.
"The pitch was off a little bit last week - it wasn't just a fluke that 20 wickets fell on a day.
"But I don't think they'll be defensive [Saturday] - I don't think they've got anything to lose."
