Tuffey, who played 26 Tests and 94 One-Day matches for the Kiwis between 2000 and 2010, is one of a number of high profile players lining up in third edition of the 16-team competition, which begins on Saturday.
The competition is divided into eight Sydney Sixers and eight Sydney Thunder conference teams, with the semi-finals and final of the high profile Twenty20 tournament to be played at the SCG on December 10.
Other key players in the Regional Bash include Adelaide Strikers batsman Jono Dean, who will be lining up for the ACT Aces, and former NSW allrounder Simon Keen, who will represent the Central Coast Rush.
The Plan B Regional Bash has a new round-robin format which replaced the previous knockout competition.
Most teams will play triple-headers over the course of a weekend before proceeding directly to the semi-finals and final at the SCG.
The tournament begins on Saturday with Sixers conference teams the Hunter Hitmen meeting the Illawarra Flames clashing at Robins Oval, Maitland and the Newcastle Blasters and the Central Coast Rush meeting at Lorn Park, Maitland. Both matches are 2pm starts.
The Thunder Conference will kick off at 2pm the following Saturday, 22 October, in a match between Riverina rivals Border Bullets, featuring Tuffey, and the Wagga Wagga Sloggers at Bilson Park, Albury.
This will be followed by the match between ACT Aces and the South Coast Crew at 9:30am on Saturday 29 October at Manuka Oval, Canberra, with all other fixtures triple-headers.
