Rugby batted the entirety of the day, finishing at 9/261, with Karydis’ 96 the standout performance.
The effort with the bat has given Rugby the opportunity to score a much-needed big win, with the men in sky blue currently last on the ladder and five points off third place.
Rugby lost Glenn Shepherd (8) early on but Lachie Harper (31) and Campbell Rose (19) rebuilt before Karydis entered the match.
Newtown, led by Mike Jeffrey (3/89) and Nathan Merchant (3/68), kept chipping away but Karydis enjoyed the moment of passing 50 as he was batting with his cousin Nick Karydis at the time.
The latter fell for 17 but the set Karydis kept pushing towards the triple figures milestone.
His knock, which included 10 fours, came to an end on 96 when trapped in front by Chris Beatty (1/23).
Mitch Russo (25) and Blake Watmore (25 not out) also chipped in as Rugby finished at 9/261 at stumps.
South Dubbo finished day one of its clash with ladder leaders RSL-Colts on top, trailing by just 96 runs.
Jeff Shanks claimed an outstanding 3/25 from nine overs as Colts was dismissed for just 135 after winning the toss and batting at Lady Cutler 4.
Colts had got to 3/97 at one stage but then collapsed, losing seven wickets for just 38 runs. Lyndon Whitney’s 36 was the top score while Nathan Finlay’s 30 was the only other score of note.
A number of Souths bowlers shone, with Tom Mitchell taking 2/25 from 10 while Andrew Cusack claimed 1/23 from 9.5 overs.
By the time Colts was dismissed there was still a tricky 17 overs remaining in the day but the Hornets saw it through unscathed.
Brad Haling and Pat Brandon both finished unbeaten on 19 and Souths will resume the chase next week well-placed at 0/39.
Shane Groen bowled a miserly 0/3 from four overs and will be key next week if Colts are to salvage anything from the match.
Three late wickets gave Macquarie a huge boost on Saturday and left its match with CYMS right in the balance.
The Blues battled early on Saturday after winning the toss and batting, being reduced to 9/69 before a potentially vital tenth-wicket partnership between Shane Dupille (19) and Waseem Khan (12) helped the score on to 102.
Andy Blake (2/14) and Brant Mann (1/4) then struck to have the Cougars 3/20 at stumps.
Earlier, Luke Patis (21) had been the only player to reach 20 as Cougars leg spinner Josh Thompson (3/18 from 12) led the way while his father Darryl (2/7) also chipped in.
But Macquarie’s bowlers also impressed, with Cal Giffin (1), Heath Larance (4) and Gavin Weekes (10) all departing. Nathan Hawke (2 not out) and Tom Coady (0 not out) will resume the chase next weekend.
