The match is a top three battle with the Colts hitting top form after their opening round loss to CYMS.
The Hornets come into the match after suffering their first loss of the season last time out against Newtown.
Colts' march up the ladder has coincided with Greg Buckley's return and the Englishman has added plenty with both bat and ball.
Coming off a star performance against Macquarie, Buckley will be vital again but will face a stern test against South's bumper batting and bowling lineup.
Despite misfiring last round, the Hornets batting lineup is the envy of much of the competition with the likes of Mitch Bower, Will Lindsay, Greg Rummans and Tim Berry all capable of big scores.
As well as those four, Henry Railz has come on in leaps and bounds this season with the young all-rounder already becoming a key figure for his side.
Bouncing back from last week's disappointing total of just 146 will be the Hornets' main target this round.
Six of the wickets in that innings fell to the off-spin of Steve Skinner and the slow bowling is again set to be a feature in this match.
Colts feature the likes of Buckley, Marty Jeffrey and Wes Giddings and they will be looking to replicate Skinner's effort.
Buckley took five wickets last week against Macquarie while Giddings starred with six the previous round.
Leg-spinner Jeffrey is also coming off a strong debut campaign with Western Zone and the three tweakers are all set to get through plenty of overs.
Colts had their bowlers to thank last week after they were rolled for just 88 on a wet deck.
They still managed to win, dismissing the Blues for 80, and then the defending premiers looked much more at ease at the crease in the second innings.
Colts piled on 297 with Buckley, Giddings and Jason Ryan all getting to spend some quality time in the middle.
Along with the likes of Paul Hulthen, Matt Keenan and captain Chris Morton, the men in red have the kind of batting depth which sets up a bumper clash with South's quality bowling unit.
