Colts brushed aside Rugby last time out but captain Chris Morton expects a much sterner test against a Blues side coming off an equally impressive win against Newtown.
But despite Macquarie's promising showing, Morton said his side is now well placed to make two wins in a row.
"We had a very convincing win and now it's time to build on that and we're close to full strength too so that helps," he said, before admitting it was a struggle during the opening rounds with so many players unavailable.
"It was very painful for the first three weeks and not just for me picking a side but second grade too.
"But having (Greg) Buckley now helps a lot and we're still missing the Cox boys but part from that we're full strength and now we can just build on what we've been doing."
Macquarie's win last round saw them move up to second on the ladder and Morton is wary of the Blues powerful batting lineup.
Both Keiran Brien and Jason Green made centuries against the Tigers but the likes of Ed Haylock, Justin Gavin and Dan Medway also capable of piling on the runs.
"They've got blokes with the bat who can really destroy you," Morton said.
"If you focus on just one or two then there's others that can hurt you and obviously 'Dicko' (Jeremy Dickson) is back and bowling well."
Dickson and Ben Taylor have formed an impressive new ball partnership for the Blues early on this season.
Morton said that is another area of concern for his side while also hoping his own opening bowlers can really fire this round.
"I think bowling was what lacked a bit for them last few years but now they've got 'Turbo' and 'Dicko' is back and I'm not sure if he's bowling as quick as he used to but he's obviously bowling good areas and taking a few wickets," he said, before speaking about his own side's bowling and fielding.
"Our intensity in the field has been fairly ordinary and our new ball bowling is not where it needs to be but our spinners do the job for us."
The spin trio of Buckley, Marty Jeffrey and Wes Giddings will again be crucial for Colts but Jeffrey will miss day two of the match due to Western Zone duties.
