Jefferies tore through the Colts line-up and finished with 5-28 from eight overs as the defending premiers rolled the men in red for just 85.
Opener Mike Deverell made 19 for Colts and that was the top score in the disappointing innings.
Rugby's Chris Wood started the run chase positively and remained at the crease right until the end, finishing 43 not out.
Ryan Davies toiled hard for Colts and took 2-21 from 7.1 overs.
Despite the emphatic win it didn't change either side's position on the ladder with Rugby second and Colts remaining third.
RSL-Colts Youth Development recorded just their second win of the season on Saturday as they edged past South Dubbo in a thriller.
Batting first, Colts YD posted 178 overs with Glenn Armstrong (56) and Garry Ashford's (31) 92-run partnership setting up the imposing total.
Ben Madigan, who had a stellar all-round match for Souths, was the best with the ball as he claimed 4-34 while Mathew Wakely chipped in with 3-30.
Souths were in trouble early during the run chase as they fell to 4-34 but a 101-run partnership between Madigan (78) and Scott Dwarte (49) got the innings back on track.
At 4-135 the Hornets looked on track but Dwarte's dismissal triggered a collapse and with the required run rate grew higher.
Souths eventually finished at 9-172 from their 40 overs, giving the predominately young Colts side a six-run win.
Sam Bass and Mike Parrish were standouts with the ball and took three wickets each for Colts YD.
Newtown Black solidified their finals position with a 54-run win over CYMS at Bob Dowling 2.
Matt Ball scored 90 not out in a man of the match effort as Newtown Black made 172 and they then dismissed the Cougars for 118.
After electing to bat after winning the toss, Ball played a lone hand for the Black Tigers with Tom Skinner's 23 the next best score.
Jason Root and James 'Busta' Nelson claimed two wickets each for the Cougars.
CYMS were steady in the run chase and were 2-63 at one stage but no one could make the big score required and Jerose Joseph's 24 the top score behind the 28 sundries.
Adam Wheeler and Peta Powyer claimed to wickets each in the routine win.
Newtown Gold enhanced their title credentials with a 97-run mauling of Macquarie at John McGrath 2.
Half centuries to Craig Pettit (56) and Mitch Gray (51) led Newtown Gold to 6-232 before Darren Davies' five wicket haul helped roll the Blues for 135.
Dylan Wilkinson made 53 in the run chase but he was the only one who could handle Davis as the veteran finished with the hugely impressive figures of 5-10 from five overs.
Earlier, Pettit and Gray had starred with the bat while there were also contributions from Jackson Dawe (37) and Sam Carolan-Work (21).
Luke Carmen took 3-24 for Macquarie but no Blues batsmen apart from Wilkinson could then get going in the run chase.
