Dubbo City Council announced the changed traffic flows would be in place until December as it reported significant progress had been made in the redevelopment of the precinct.
It was about to start the third stage of the project to replace the degraded eastern footpath, kerb and guttering, and road pavement along the street's shoulders between Wingewarra Street and Talbragar Street.
Council works services manager Ian Bailey said new stormwater pipes had been laid and the London plane trees removed in the first two stages of the project.
"Stage three does require the closure of Darling Street to southbound traffic to enable the major construction work to commence," Mr Bailey said.
From Sunday, traffic management conditions in place between Wingewarra Street and Talbragar Street include the closure of the southbound lane of Darling Street until further notice.
One northbound lane would be open under traffic management conditions, which Mr Bailey said were designed to provide continued access to Dubbo Public School, Darling Street businesses and Dubbo Regional Theatre and Convention Centre.
"Motorists will be able to travel north from Wingewarra Street to Talbragar Street with parking and drop off zones unaffected on the western side of Darling Street," Mr Bailey said.
The Darling Street gates of Victoria Park Number One Oval would also be closed to all vehicles, with access to the oval from its eastern gates and parking available at the northern end of Darling Street.
Pedestrian access from Number One Oval to Darling Street will be maintained via a dedicated pedestrian gate at the Darling Street entrance gates.
"It is expected that these arrangements will be in place until December this year and motorists are encouraged to use alternative routes via Brisbane Street or Gipps Street if travelling south," Mr Bailey said.
